3.  Replacement vector for targeted disruption of the murine HCII gene

The blue boxes represent exons 1-4 of the HCII gene.  The thick lines represent genomic DNA; the thin lines, vector DNA.  The neomycin phosphotransferase gene (neo) was inserted in place of ~1 kb of 5' flanking sequence, exon 1, and part of intron 1.

The probe used for Southern blotting hybridizes with a sequence in exon 4.  Digestion with SacI yields a 14-kb DNA fragment from the wild-type locus and a 6-kb fragment from the targeted locus.

RW-4 embryonic stem cells derived from 120/SvJ mice were transfected with the linearized targeting vector and selected for growth in the presence of G418.
