4.  Disruption of the HCII gene in mice

Embryonic stem cells were injected into C57Bl/6 blastocysts and implanted into pseudopregnant females.  The resulting chimeric males were crossed with wild-type C57Bl/6 females to produce F1 offspring.  Heterozygous HCII+/- mice were crossed to produce HCII-deficient mice.

A.  Southern blots of genomic DNA digested with SacI.

B.  Western blots of plasma probed with goat anti-human HCII IgG (Affinity Biologicals, Hamilton, Ontario).  The 68 kDa and 72 kDa bands represent the two glycoforms of HCII present in mouse plasma (Zhang et al., Biochemistry 33: 3632, 1994).
