8.  Plasma antithrombin activity

Antithrombin activity in mouse plasma was determined by measuring the amount of human factor Xa inhibited in the presence of heparin.

Reaction mixtures (100 µl final volume) contained the following:

Mouse plasma (0.3 µl)
Heparin (1 unit/ml = 7 µg/ml)
Human factor Xa (11 nM)
After 1 minute, the amount of residual factor Xa activity was determined by hydrolysis of a chromogenic substrate.  The antithrombin activity is expressed as the mean + 1 SD (n = 9-11 animals of each genotype).
