Antithrombin activity in mouse plasma was determined by measuring the amount of human factor Xa inhibited in the presence of heparin.
Reaction mixtures (100 µl final volume) contained the following:
Mouse plasma (0.3 µl)After 1 minute, the amount of residual factor Xa activity was determined by hydrolysis of a chromogenic substrate. The antithrombin activity is expressed as the mean + 1 SD (n = 9-11 animals of each genotype).
Heparin (1 unit/ml = 7 µg/ml)
Human factor Xa (11 nM)
