4. Thrombin time of purified fibrinogenInitially, we considered the possibility that the monoclonal IgG interferes with the ability of thrombin to cleave fibrinogen. As shown below, the IgG did not prolong the clotting time of purified human fibrinogen. [Previously, we found that much higher concentrations of the IgG (>130 µM) prolonged the reptilase time of plasma, apparently by interfering with fibrin monomer polymerization.]
Addition of the monoclonal IgG to a mixture of fibrinogen and antithrombin, each present at approximately its normal plasma concentration, prolonged the thrombin time to an extent similar to that observed after addition of the IgG to plasma.
