We have reported that the patient's monoclonal IgG binds to human alpha-thrombin, as well as to beta- and gamma-thrombin. The IgG does not bind to prothrombin, to other vitamin K-dependent coagulation factors, or to fibrinogen. The observation that the IgG does not bind to prothrombin is consistent with binding of the IgG to exosite II of thrombin. In prothrombin, exosite II is covered by the fragment 2 domain and is inaccessible to the IgG.
